Continuous-Flow Magnetic Fractionation of Red Blood Cells Based on Hemoglobin Content and Oxygen Saturation—Clinical Blood Supply Implications and Sickle Cell Anemia Treatment

Approximately 36,000 units of red blood cells (RBCs) are used every day in the U.S. and there is a great challenge for hospitals to maintain reliable supply, given 42-day expiration period from donation date. For many years, research has been conducted develop ex vivo storage solutions that limit RBC lysis high survival rate transfused cells. However, little attention directed towards potential fractionation methods remove unwanted cell debris or aged stored prior transfusion, which could not only expand shelf life but also avoid adverse events patients. Such number transfusions required treating certain pathologies, such as sickle disease (SCD). In this work, magnetic studied technology fractionate functional healthy RBCs It reported during storage, lose hemoglobin (Hb) lipid content via formation Hb-containing exosomes. Given character deoxygenated- met-Hb, we propose use quadrupole sorter (QMS) based on their Hb both SCD blood. our QMS, cylindrical microchannel placed inside center quadrupolar magnets subjected fields constant field gradients (286 T/m), causes deflection paramagnetic, Hb-enriched, original path collection into different outlet. Our results demonstrated although obtain significant difference mobility sorted fractions (corresponding more than 1 pg per cell), exists tradeoff between throughput purity.
